I think the curse came after invading the land of the giants. My theory on this is that the curse is want; desire. I believe Nashandra manipulated Vendrick into invading the land of giants for the petrified dragon egg, knowing the ancient dragons lacked the curse of want, which would be the antithesis for what is required to spread the darkness. She wanted to keep the egg, and Vendrick close, but Vendrick bucked. I think Vendrick became aware of the cycle, and after working on a way to stave off the curse, he set things up to keep Nashandra away from the important places/things. Himself included.
More speculatively, I think the giants were created from trees the way the golems were given life. I don't think the power of the giants was a power they controlled themselves, but were given by whoever created them. To go a little further, I believe they were created from archtrees by the dragons as guardians, which is why they're "legion", and why they are almost mindless in their attempt to retrieve what Vendrick stole. It would also explain why they turned to tree upon being killed. In short, I don't think the giants wield a great power so much as their very existence was built upon a great power.
I think Vendrick lost all hope when he realized that "the curse of life is the curse of want", and that the curse could not be "cured" so long as man is man. He failed in creating a human/dragon hybrid, and the rest of the big players were consumed by their desire, to disastrous effect.
I don't think there is a good ending to be had. I believe stoking the first flame is the best case scenario, because it's either the cycle, or the darkness. I believe the curse was born out of the lighting of the first flame by Gwyn, and that as long as their is the flame, there is the curse. It was all born of desire, and man is to pay for that desire as long as man exists.
Man is desire; there's no separating the two, in essence. I believe this is what Vendrick saw when he peered into the very essence of the soul.

Look at the NPCs in Majula. Every single one talks about what they want and why they wanted to come there, but none of them remember anything else. The armor salesman, for instance. He becomes rich and then forgets everything else. I think you're right... the game is about greed.

Great post here, but I have one question. How did Vendrick fail in creating a human/dragon hybrid?

The Emerald Herald, Shanalotte: Born of dragons, contrived by men. She says she came out wrong, and I believe this to mean that she was created in the hopes that she could not be cursed (due to her having been born of dragons,) but ended up cursed anyway.
I think she may have been created to light the first flame, as someone who was free from the curse of want, in the hopes that this may end the cycle and the curse. Maybe this is why the Lost Sinner punishes herself, because she came to understand that she would only have perpetuated the cycle somehow.

I don't think Shanalotte is cursed because she remains lucid throughout the entire game regardless of the fact that you are (in a roundabout way) giving her thousands of souls.
Also it's strange that Maughlin becomes less and less sane the more you buy from him; I thought souls cured the ailing mind?

He also gets more armour the more you pay him. He may not know his hometown but he knows how to invest in his business.
